Gene Synthesis - FAQ

We guarantee 100% accuracy by sequencing. You will receive complete QC data. Each gene comes with complete QC data: project report, construct map, complete sequence, sequence verifying data (chromatograms) and alignment file.

We deliver 4ug of lyophilized plasmid, which contains your target gene in our standard vector or in your desired vector. We can prepare more plasmid (up to a few milligrams) upon request.

The plasmid is shipped at room temperature. Our typical turnaround time is 2-3 weeks for a standard gene under 2-3 kb. The turn-around time varies depending on the gene length, subcloning and difficulty.

Although gene synthesis method has no theoretical limits, longer genes require extra planning and have a longer turnaround time and can sometimes be very difficult to synthesize. We encourage our customers to synthesize no more than 10kb in length. It costs much more, and take longer time to synthesize longer genes than 10kb.

We have extensive experience in synthesizing difficult genes. Our synthesis platform allows us to produce very complex genes with high GC content, repetitive elements, or wobble bases.

Our method is very reliable across a large range of AT to GC ratios. We have already synthesized genes with a GC content of over 85% successfully. If, however, your gene has a high GC content of >70%, and additional critical features such as many repetitive elements, you should contact our support team prior to ordering a gene.

Expression in different organisms or tissue types is often enhanced by the use of a special subset of codons. For many organisms and tissues, codon usage tables have been computed from genes with high expression in these target organisms and tissues. By adjusting the codon usage of your synthetic gene to the codon usage of your host organism, you will likely increase its expression efficiency, and consequently, the yield of the expressed protein. We can also optimize a gene using a mixed codon table computed from the codon usage tables of the two organisms.

Yes, you can. We offer this service for completely degenerated positions (all four nucleotides in roughly the same amount) without an additional fee. Ask for more complex degenerations (e.g. only A and T) - they are also possible without any problems. Please note that the exact ratio of all bases at a degenerated position cannot be predicted.

Peptide Synthesis - FAQ

Our typical turnaround time is 2-3 weeks for a standard purified peptide under 30 amino acids. The turnaround time varies depending on the peptide length, solubility, and difficulty. Turnaround Time (TAT) indicated on the quote is an estimated time only. It does not reflect any production difficulties, if so encountered. All peptides will be shipped in lyophilized powder, at room temperature.

By default, each peptide comes with CoA, MS spectra, and HPLC chromatograms. See Sample QA Report. We can provide additional analysis if required, e.g. Peptide content analysis, TFA content analysis, and KF test (Water content test).

Biomatik can synthesize peptides up to 130aa. Unlike many peptide suppliers who are only comfortable in making peptides under 30 or 40aa, Biomatik has extensive experience in making peptides ranging from 40aa to 90aa. However, the success rate is getting lower when it comes to longer peptides, especially 100aa or longer. If you plan to synthesize a sequence of 130aa or longer, please contact us for custom protein expression and purification service.

Each peptide has its specific characteristics. If some problems happen during the synthesis beyond our expectation, and we cannot deliver your peptide on time, we will inform you as soon as possible. By chance that we are not able to make the peptide, you will not be charged for any costs - which is our "No Peptide, No Charge" policy.

The sequence, amino acid composition, and length of a peptide will influence whether correct assembly and purification are feasible. These factors also determine the solubility of the final product. Please refer to our Peptide Design Guideline on some factors in the design of a peptide sequence for synthesis.

Please refer to our Recommended Peptide Purity Guideline.

By default, all research peptides are synthesized in TFA salt form. For cell-based assays or animal studies, you should consider having TFA salt removed by switching it to acetate or HCl salt form (with TFA <1% guarantee) to avoid abnormal responses. Depending on the budget, you may also want to consider higher purities (>98%) to get optimal results.

Peptide solubility information is included on the CoA if you have requested a solubility test at the time of ordering, free of charge. If it is not available, you can follow a Peptide Handling Guideline.

For short peptides with normal sequences under 15aa, it is generally 40-60% by HPLC for crude grade; 50-70% by HPLC for desalted grade. The longer the peptide, the lower the purity for crude or desalted. Peptides are generally purified by HPLC using water and acetonitrile gradient. Most impurities are fragments or deletion peptides, incompletely de-protected peptides, and residual salt and water.

Research grade custom peptides come with limited quality control (MS and HPLC analysis only). Therefore, the variation is expected from batch to batch in terms of the net peptide content of the desired target peptide, salt content, water content, solubility, etc. Such batch-to-batch variation shall not create an issue for most experiments.

However, if you are engaged in sensitive bioassays and you want to minimize batch-to-batch variation or determine the exact peptide dosage used in the experiments, you shall request additional quality control and analysis (e.g., peptide content, salt content, water content, solubility), at an additional cost.

For example, if you are engaged in cell related assays, you shall request TFA removal (<1%); If you must apply the same peptide dosage for your experiment from batch to batch, you shall require peptide content analysis; If you need high solubility or concentration (e.g. 5mg/ml) in a specific solvent from batch to batch, you shall ask for solubility control and testing. For the ultimate batch-to-batch consistency, consider GMP-grade peptides.

Recombinant Protein Synthesis - FAQ

We offer protein expression in:

a. Bacterial systems: Cost-effective and fast production for non-glycosylated proteins.
b. Mammalian systems: Best for human-like glycosylation and authentic post-translational modifications.

Yes, we offer a risk-free guarantee for proteins between 10-60 kDa: no protein, no charge. For proteins smaller than 10 kDa or larger than 60 kDa, only the gene synthesis and project setup costs will be charged if the project is unsuccessful.

We have a success rate of over 95% for protein production using the E. coli expression system. However, we do not guarantee protein activity or suitability for downstream applications.

Our comprehensive service includes:
• Codon optimization and gene synthesis
• Vector design and cloning
• Small-scale test expression
• Protein purification and quality control
• Scale-up production

Turnaround times vary depending on the protein and expression system. Generally, bacterial expression projects take 4-6 weeks, while yeast, insect, and mammalian systems may require 6-12 weeks. We provide project-specific timelines after an initial assessment.

Yes, we can scale up production to milligrams or grams of protein. We typically start with flask culture to determine the yield through a pilot fermentation study. If the yield is sufficient, scaling up to larger quantities is possible.

Antibody Production - FAQ

We offer recombinant antibody production, Single B cell monoclonal antibody production, and polyclonal antibody production.

Yes, we offer some of the best guarantees in the industry. Contact us for details or download our antibody package information from our website.

Peptide Length: Typically 12–22 amino acids; avoid peptides <8 amino acids unless necessary.

Carrier Protein: KLH is preferred; other carriers like albumin are also available.

Conjugation Chemistry: We recommend sulfhydryl cross-linkers for peptides without cysteine.

We require 10 mg of protein (>90% purity) in two 5 mg sets. Provide SDS-PAGE data and ship as lyophilized powder (preferred) or neutral buffer (>1 mg/mL).

Yes, we provide free peptide design with antibody projects. Our team analyzes your protein sequence for the most immunogenic regions and offers recommendations.

Species and numbers depend on the antigen and required serum volume. Typically, two rabbits are used, but for specific needs, goats, chickens, rats, mice, and guinea pigs may be recommended.

Yes, but mixing peptides in the same immunization may create competition. Separate immunizations are recommended for distinct peptides.

Custom antibodies are typically shipped as lyophilized powder.

Catalog ELISA Kits - FAQ

Components in the kits either need to be stored cold (2-8°C) or frozen (-20°C). Different kits may have different storage conditions, please check the indicated storage conditions upon receiving.

Dilution for reagent A and B does not necessarily have to be done all at once. It is recommended that they be kept in their stock solution to maintain stability. Only prepare as much reagent as you intend to use for that immediate experiment. The reconstitution standard, however, cannot be kept as it degrades after storage at low concentrations. It is recommended to only use the reconstitution standard once.

Finish preparing the reagents 10 minutes before use. Concentrate all reagents to the bottom of the tube through centrifugation when using for the first time. Use precise measuring tools to ensure the most accurate preparation (pipette, graduated cylinder etc.). NEVER assume the quantity by adding directly to the supplied vials. Often, the amount in the tube is larger than specified. Ensure all labwear is clean before beginning.

Pre-coated plate, standards, detection reagent A, and detection reagent B are core reagents in the kit. If you are concerned about the kit quality after transit, you can run one or both of the following tests. If you are looking to conserve reagents and only validate TMB and detection buffer B, try Test #1. If you wish to validate the whole kit, run Test #2 or both Test #1 and #2.

●Test #1: This test is used to validate the TMB substrate and detection B after transportation. Draw 100ul of TMB substrate and place in a clean EP tube, this should be colorless. Using a new pipette, add in 1ul of the detection reagent B. Mix well and observe for color. If there is color present, it means that the TMB substrate and detection B are good - this is a good indicator that the whole kit is good after transit.

●Test #2: This test is used to validate the whole kit (validate all reagents). Run a pilot experiment with different dilutions of the standard with one or two samples to test. The standard curve can be used to determine whether the whole kit is still good or not, and it can also be served as the positive control and negative control both.
