• Biomatik's tag-free protein production is unlike the traditional tag removal process. Our tag-free protein starts with tag-free protein expression, and purified by SEC and ion exchange to ensure proper native folding. Customers receive high quality protein with no trace of the affinity tag since no tag removal involved in production.
• If the expressed protein is insoluble (expressed as inclusion body), refolding will be attempted to make it soluble.
• Protein purity is determined by SDS-PAGE, and quantity is determined by Bradford assay.
• Protein is delivered in freeze powder whenever possible (in neutral buffer before lyophilization: 1x PBS, pH 7.4). If protein is delivered in solution form, the default buffer is: 1 x PBS, 50% Glycerol, pH 7.4.

The conventional recombinant protein production introduces different tags (typically His-tag or GST-tag) to carry out protein expression and purification. Unfortunately, these tags cannot all be removed and can create real issues in downstream applications. Unlike traditional post-production tag removal, Biomatik's tag-free protein production service ensures proper native folding, and no trace of the affinity tag (no tag cleavages). Endotoxin Removal Service from Biomatik
Endotoxins are associated with the outer cell membrane of gram-negative bacteria such as E. coli. The major component of the outer cell wall membrane is composed of Lipopolysaccharides (LPS) which can elicit a range of inflammatory responses in the mammalian hosts and can often be detrimental to such host systems. It is necessary to remove these endotoxins from the final protein preparation for downstream applications involving in vivo studies or cell-based assays. Biomatik’s Endotoxin Removal Service can be added as part of your custom protein expression service, and we can reduce the endotoxin level to below 1EU/ug with great success.

1. What is the typical turn-around time for custom protein productions?
Our typical turn-around time is 6-8 weeks for custom protein productions. The turn-around time could vary depending on the complexity of the protein properties and if additional service such as endotoxin removal is requested.
2. How do you ship and how long does it take to reach me?
Recombinant proteins are typically delivered in lyophilized format (unless otherwise requested) and shipped on dry ice. The shipment is sent by overnight delivery within USA/Canada, and may take 3-4 days to reach researchers in other countries.
3. What QC data do you provide and how do you determine purity?
Quality assessment and purity determination of the custom protein is done by SDS-PAGE/Coomassie staining. His-tag proteins will also be assessed by western-blot analysis, while tag-free proteins can be assessed by optional LC-MS-MS analysis.
4. What if the protein expression construct does not express well?
On rare occasions, despite our optimization efforts, the plasmid construct does not yield optimal protein expression level on the first attempt. There are several approaches we may take to troubleshoot; we may re-clone the gene sequence into a different expression vector, we may use different E. coli strains, or we may optimize the induction/expression conditions. Rest assured, we will not give up until all options have been explored which accounts for our high success rate.
5. I will be using the recombinant protein for antibody production, are there any specific requirements I need to consider for my custom protein production?
- The protein antigen should be dissolved in neutral buffer such as PBS.
- His-tag is the preferred tag due to its small size and less likely to elicit background response.
- Higher purity such as >90% is recommended.
